摘 要:对蛹虫草(Cordyceps militaris)原生质体分别进行热应激和紫外线诱变处理后,挑选再生菌株进行栽培试验,通过测定菌株子实体产量及腺苷、虫草素含量,从中筛选高产突变菌株。蛹虫草原生质体经68℃和72℃热应激后,再生菌株子实体干物质含量显著增加,且68℃处理组子实体中腺苷含量显著提高。用紫外灯照射原生质体1 min、6 min和7 min时,子实体干物质含量显著增加,腺苷含量较对照组分别提高49.4%、96.4%和15.9%,虫草素含量分别提高49.1%、20.0%和13.5%。最终从紫外线诱变1 min处理组筛选出5株高产突变菌株,其子实体干质量和生物转化率均显著高于出发原菌株,子实体中虫草素含量分别为原菌株的1.59、1.72、1.74、1.55和1.56倍,并且5株菌株目标性状的稳定性良好。试验结果表明,以原生质体为材料,采用紫外线诱变技术可以选育出蛹虫草高产突变菌株。Regenerated strains of Cordyceps militaris after heat stress or UV mutagenesis of protoplast were cultivated by fruiting test. The productivity,adenosine and cordycepin content of fruit bodies were determined. Mutant strains with excellent comprehensive properties were screened out. Dry matter content of fruit body was improved significantly by 68 ℃and 72 ℃ heat stress treatments of protoplast,and adenosine level increased significantly in 68 ℃ treatment group. After protoplast exposure to UV for 1 min,6 min and 7 min,dry matter content in fruit body of the mutant strain was improved significantly; adenosine content was increased by 49. 4%,96. 4% and 15. 9%,respectively; cordycepin content was increased by 49. 1%,20. 0% and 13. 5%,respectively. Five high yield mutant strains with higher performance were screened out from the 1 min UV treatment group. The dry matter of fruit bodies and biological conversion rate were significantly higher than the original strain. The cordycepin content in these five strains was 1. 59,1. 72,1. 74,1. 55 and 1. 56 times to that in the original strain,respectively. Besides,target characters of these 5 strains were stable. These results indicated that UV mutagenesis of protoplast could obtain high yield mutant strains for Cordyceps militaris cultivation.
