摘 要:为了探讨鹅血有效蛋白组分对胃癌细胞增殖的影响,并通过质谱鉴定初步了解鹅血有效组分的多肽和蛋白组成及其功能。取鹅血清,并分级分离鹅血有效成分,再向HGC-27和SGC-7901细胞中分别加入各组鹅血蛋白组分、阳性对照化疗药5-FU、空白对照作用36 h,用MTS法检测细胞活力。最后,对有效鹅血蛋白组分溶液内酶解20 h后,采集和分析提取出的酶解多肽,质谱鉴定有效鹅血蛋白组分的构成。结果显示,30 kD以下组分和10 kD以下组分可明显抑制HGC-2细胞和SGC-7901细胞活力,其中10 kD以下组分的抑制作用更明显。然而,3 kD以下组分对细胞活力的抑制作用较弱。本研究还发现稀释10倍后的10 kD组分对细胞活力的抑制作用不明显。接着,本研究还对鹅血清中10 kD组分进行了质谱分析,共获得135个蛋白或多肽,其中功能已知的蛋白为60个,与其他物种有类似功能结构域的蛋白48个和未知蛋白27个,其中5个多肽初步分析可能参与机体的防御反应。因此,本研究初步认为鹅血清的蛋白组分具有一定的抑制胃癌肿瘤细胞生长的作用,其主要成分集中于10 kD组分。To explore the effects of effective protein component of goose blood on the proliferation of gastric cancer cell,and to have a preliminary understanding about the composition and function of the effective components of goose blood through the mass spectrometry.Take some goose serum,and fractionate the effective ingredients of goose blood.Treatment of gastric cancer cell lines HGC-27 and SGC-7901 with each group goose blood protein components and positive control drug 5-FU for 36 h and using MTS assay to detect cell viability.Get the effective components and conduct an enzyme of solution for 20 h and then extract the enzyme solution peptides.Subsequently,use mass spectrum to collect and analyze,and identify the composition of effective goose blood protein components.30 kD and 10 kD components both significantly reduced cell viability of gastric cancer cell lines HGC-27 and SGC-7901 and 10 kD components with more obvious inhibition effect.But 3 kD components had no significant inhibition on the viability of HGC-27 and SGC-7901 cells.Our results also showedthat after 10 kD components were diluted 10 times,its inhibition effect on viability of HGC-27 and SGC-7901 cells was not obvious,which suggested that this action is dose-dependent.Furthermore,we also used mass spectrometry to detect 10 kD components of goose serum to further understand the protein components which may work.The results of the mass spectrum analysis showed that 135 proteins or peptides were obtained.Among them,there were 60 proteins whose function had been known,48 proteins which had have a similar function structural domain with other species,and 27 unknown proteins.The sequence mainly referenced the genetic information of the similar species anas platyrhynchos.The number of amino acids of all the proteins is was between 43 and 3201.Among them,there were 19 proteins or polypeptides which had less than 130 amino acids.By preliminary analysis,5 polypeptides in the components may be involved in the defense reaction.Goose serum protein components have
