摘 要:研究NO调节采后番茄果实产生抗病反应的机制中MAPK和乙烯的作用。采用0.1 mmol/L一氧化氮供体硝普纳(SNP)和0.1 mmol/L SNP与10μmol/L MAPK抑制剂1,4-二氨基-2.3-二氰基-双氨基苯巯基二丁烯(U0126)结合处理的方式处理采后普通丽春番茄果实和转反义ACS2番茄果实。结果表明:0.1 mmol/L SNP促进了处理后0~24 h内的普通丽春番茄果实中SlMAPK1/2/3的表达,刺激了内源乙烯合成关键基因SlACS2/4的表达和1-氨基环丙烯-1-羧酸合成酶(ACS)的活性,诱导内源乙烯释放量增加,同时刺激了番茄果实苯丙氨酸解氨酶(PAL)和几丁质酶(CHI)活性的上升;而该效应可被10μmol/L U0126抑制。转反义ACS2番茄果实中SlMAPK1/2/3的表达也受到SNP的刺激,然而乙烯合成和PAL以及CHI不受SNP处理的影响。结果表明SlMAPK1/2/3和SlACS2均参与SNP对采后番茄果实抗病酶PAL和CHI的诱导,SlMAPK1/2/3在乙烯上游。This sduty pay attention to the effect of Mitogen-activated protein kinase(MAPK) gene SlMAPK1/2/3 and SlACS2 in NO-induced phenylalnine ammonialyase and chitinase of postharvest tomato fruits. Experiment treatmented the postharvest Lichun tomato and antisense-ACS2 tomato fruits by 0.1 mmol/L nitric oxide donor sodium nitroprusside(SNP)and(SNP +10 μmol/L inhibitor of MAPK 1,4-diamino-2,3-dicyano-1,4-bis(o-amino-phenyl-mercapto) butadiene(U0126)). The results showed that in the Linchun tomato fruits 0.1 mmol/L SNP stimulated the expression of SlMAPK1/2/3 and 1-aminocyclopropane-1-carboxylic acid synthase(ACS) gene SlACS2/4, the activity of ACS, the ethylene production, and the activity of phenylalnine ammonialyase(PAL) and chitinase(CHI), and 10 μmol/L U0126 inhibited the effect of SNP, while in the antisense-ACS2 tomato 0.1 mmol/L SNP stimulated the expression of SlMAPK1/2/3, but does not affect the ethylene production and the activity of PAL and CHI. Above results suggested that SlMAPK1/2/3 and SlACS2 involved in NO regulation of PAL and CHI in tomato fruits and SlMAPK1/2/3 located upstream of ethylene.
