摘 要:以Fe^(2+)和Fe^(3+)制备的磁性纳米粒子,通过X-射线衍射测定其晶格、傅里叶变换红外光谱测定其非极性化学键确定合成的磁性纳米粒子为Fe_3O_4、扫描电镜结果表明转速为100 r/min磁性纳米粒子形貌不统一、200 r/min磁性纳米粒子呈椭球形。将伴刀豆蛋白A分别修饰到两种磁性纳米粒子表面,激光粒度仪度分别测定了修饰后Fe_3O_4@Con A的平均粒径分别为(82.49±20.34)nm和(9.77±0.02)nm,振动磁强计测量结果表明修饰后Fe_3O_4@Con A磁强度分别为3.68 emu/g和5.36 emu/g。二种粒径的Fe_3O_4@Con A从脱脂乳中回收乳铁蛋白的洗脱液的十二烷基硫酸钠聚丙烯酰胺凝胶电泳(sodium dodecyl sulfate polyacrylamide gel electrophoresis,SDS-PAGE)结果表明,200 r/min制备的Fe_3O_4@Con A对脱脂乳中回收乳铁蛋白的专一性优于100 r/min的Fe_3O_4@Con A。Magnetic nanoparticles were prepared with Fe2+ and Fe3+, X-ray diffraction and Fourier transform infrared spectrometry were employed to measure the lattice and nonpolar chemical bonds to identify the magnetic nanoparticles were Fe3O4, results of scanning electron microscope showed that the magnetic nanoparticles obtained by 100 r/min with not uniform morphology and spheroidicity and uniform particle size of magnetic nanoparticles were obtained by 200 r/rain. Concanavalin A was modified on the surface of nanopariclesobtained by 100 r/min and 200 r/min, and particle size analyzer showed that the average particle size were (82.49±20.34 ) nm and (9.77±0.02 ) nm. Results of vibrating sample magnetometer demonstrated that the magnetic capacity of magnetic nanoparticles obtained by 100 r/min and 200 r/min were 3.68 emu/g and 5.36 emu/g, respectively. Modified nanopaticles were used to recover latcoferrin from skimmed milk, and the results of sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE ) demonstrated that the specifity of nanopaticles obtained by 200 r/min was better than that of 100 r/min.
