摘 要:目的研究RGS1在人黑色素瘤细胞中对PD-L1表达的调控和影响,进而探究RGS1对恶性黑色素瘤发生、发展的作用机制。方法⑴选取34组黑色素瘤临床样本,对每组标本的病理切片分别行抗RGS1和抗PD-L1免疫组化染色,并且分析在黑色素瘤细胞中,两种分子的表达是否具有相关性。⑵选取A375(人体黑色素瘤)细胞,构建RGS1过表达组、RGS1敲低组和对照组,分别测定各组细胞中PD-L1的转录和表达水平,并且分析RGS1对PD-L1是否具有调控作用。结果分别以色素痣的RGS1和PD-L1的免疫组化染色深度作为基准,将34例黑色素瘤临床样本染色分组。发现PD-L1与RGS1在人黑色素瘤细胞中的表达具有正相关性(P<0.05)。在细胞实验中,RGS1敲低组的PD-L1蛋白表达量出现明显降低;而RGS1过表达组的PD-L1蛋白表达量及mR NA表达量较对照组均出现明显增高。说明PD-L1的表达会随RGS1的表达变化而产生变化。结论在恶性黑色素瘤中,RGS1对PD-L1的表达可能具有正向调控作用。Objective To study the regulation and influence of RGS1 on PD-L1 expression in human melanoma cells, and to explore the mechanism of RGS1 on the occurrence and development of malignant melanoma. Methods(1) Thirty-four groups of melanoma samples were selected and subsequently underwent anti-RGS1 and PD-L1 immunohistochemical staining. The correlation between the expression of two molecules in melanoma cells was analyzed.(2) A375 cells(human melanoma cells) were selected to construct three groups: an RGS1 overexpression, knockdown, and control group. The transcription and expression level of PD-L1 in each group of cells was determined to analyze whether RGS1 has a regulatory effect on PD-L1. Results Based on the immunohistochemical staining depth of RGS1 and PD-L1 in pigmented nevus, 34 melanoma samples were stained and divided into 3 groups. The expression of PD-L1 and RGS1 in human melanoma cells was found to be positively correlated(P〈0.05). In the cell experiment, the expression of PD-L1 protein in the RGS1 knockdown group decreased significantly, but the expression of PD-L1 protein and mR NA in the RGS1 overexpression group was significantly higher than that in the control group. It showed that the expression of PD-L1 will change with the change of RGS1 expression.Conclusion In malignant melanoma, the expression of PD-L1 was maybe positively regulated by RGS1.
