摘 要:以灵芝子实体为原料,研究复合酶(纤维素酶、半纤维素酶、木瓜蛋白酶)提取灵芝总三萜的工艺条件。在酶解时间、酶解温度和酶解pH三个单因素试验的基础上,利用响应面法优化复合酶提取灵芝总三萜的工艺条件。酶解提取工艺条件为:酶解处理时间、温度和pH分别为90 min、50℃和5,酶解处理后的原料继续用无水乙醇80℃水浴回流提取2 h。在此条件下灵芝总三萜的提取得率为1.29%±0.04%,与理论预测值1.28%接近。复合酶提取方法简单、效率高,可以用于提取灵芝总三萜。Ganoderma lucidum fruit body was used as raw materials to study complex enzyme(cellulose, hemicellulase and papain) extraction technology of total triterpenoids from Ganoderma lucidum. Response surface methodology was used to optimize the extraction process of complex enzyme of total triterpenoids from Ganoderma lucidum based on the results of three single factors of hydrodysis time, hydrodysis temperature and pH. The optimum extraction conditions were as follows: hydrodysis time 90 min, hydrodysis temperature 50℃ and pH 5. Then the enzyme hydrodysis treatmented materials was refluxed with ethanol for 2 h in 80℃. Under these conditions the yield of total triterpenoids from Ganoderma lucidum was 1.29%±0.04% closing to predicted extraction rate 1.28%. This extraction technique was simple, efficient and applicable for the extraction of total triterpenoids from Ganoderma lucidum.
