摘  要:目的:观察针刺对SAMP8小鼠学习记忆能力和海马细胞间液乳酸、葡萄糖、丙酮酸含量的影响,探讨针刺改善学习记忆能力的神经元能量底物供应机制。方法:SAMP8小鼠随机分为模型组和针刺组,以同月龄SAMR1小鼠作为对照,每组6只。针刺组选择"百会"、"涌泉"穴进行针刺,1次/d,5次一疗程,共针刺8个疗程。以Morris水迷宫评价学习记忆能力,脑微透析系统对海马细胞间液乳酸、葡萄糖、丙酮酸进行取样和检测。结果:与对照组相比,SAMP8组小鼠逃避潜伏期显著延长(P <0. 01),目标象限游泳路程百分比显著下降(P <0. 01),穿越有效区次数显著减少(P <0. 01),海马乳酸、葡萄糖、丙酮酸含量显著降低(P <0. 01);与模型组相比,针刺能改善小鼠第4、5天逃避潜伏期(P <0. 05),提高目标象限游泳路程百分比(P <0. 05),增加穿越有效区次数(P <0. 05),显著降低海马乳酸、葡萄糖、丙酮酸含量(P <0. 01)。结论:针刺能改善SAMP8小鼠学习记忆能力,降低海马细胞间液乳酸、葡萄糖、丙酮酸含量,可能与增加底物储备、提高细胞响应的底物匹配能力有关。[ Abstract] Objective: To observe the effect of acupuncture on learning and memory ability and on the level of lactate,glucose and pyruvate in brain interstitial fluid of SAMP8, so as to explore the neuronal energy substrate supply mechanism of promoting learning and memory ability. Methods: 12 male SAMP8 mice were randomized into model group and acupuncture group, take 6 male SAMR1 mice as control group. The acupuncture treatment on DU20 ( Baihui) and KI1 (Yongquan) was applied once daily, and 5 days as a treatment course, last for 8 courses with 2 days interval between adjacent courses. The learning and memory ablility of SAMP8 and SAMR1 mice was detected by using Morris water maze (MWM) test, the level of lactate, glucose and pyruvate in brain interstitial fluid was sampled and detected by microdialysis system. Results: Compared with the control group, the escaping latency of acupuctue group was significantly prolonged ( P 〈 0. 01 ), the percentage of distance of swimming in target quadrant was significantly decreased (P 〈0. 01 ), the number of times crossing the effective area was significantly reduced (P 〈0. 01 ), the level of lactate, glucose and pyruvate in brain interstitial fluid were significantly decreased ( P 〈 0. 01) . Compared to model group,mice in acupuncture group showed an improvement in the escaping latency in 4 th and 5th day (P 〈0. 05),and in the percentage of distance of swimming in tar-get quadrant (P 〈0. 05 ), the number of times crossing effective area ( P 〈 0. 05 ), and a decrease in the level of lactate, glucoseand pyruvate in brain interstitial fluid (P 〈0. 01) . Conclusion: Acupuncture can improve the learning and memory ability of SAMP8 mice, and decrease the level of lactate, glucose and pyruvate in hippocampal brain interstitial fluid, which may be related to its effect in energy matching and reserve capacity.
