摘  要:目的:研究芡茎多糖脱蛋白最优工艺。方法:以芡茎多糖蛋白脱除率和多糖损失率为指标,考察沙维积(Sevag)法、三氯乙酸(trichloroacetic acid,TCA)法、木瓜蛋白酶法、木瓜蛋白酶-TCA法4种方法对芡茎多糖脱蛋白的效果,从而筛选得木瓜蛋白酶-TCA法为较优方法。利用响应面法对木瓜蛋白酶法脱蛋白工艺进行优化,并将最优工艺与TCA法结合处理1次。结果:木瓜蛋白酶-TCA法最优工艺为加酶量580 U·m L-1,酶解温度30℃,酶解时间2.0 h,p H7.0,TCA体积分数8.0%,在此条件下,芡茎多糖蛋白脱除率为85.1%,多糖损失率为16.3%。结论:木瓜蛋白酶-TCA法是一种较优的芡茎多糖脱蛋白方法。Objective: To optimize deproteinized technology of polysaccharide from Euryale petioles and pedicel. Methods: With the rates of deproteination and polysaccharide loss rate as indexes,four deproteination methods,including Sevag,trichloroacetic acid( TCA),papain and papain-TCA( papain hydrolysis followed by TCA precipitation) were compared in our study,the response surface methodology was performed with a Box-Behnken central compostite experiment to optimize deproteinization conditions,and the optimal process was treated with TCA once. Results: The optimal conditions were as follows: hydrolyzing substrates with papain dosage of 580 U·mL^-1,hydrolysis temperature of 30 ℃,hydrolysis time of 2.0 h,pH of 7.0,the concentration of TCA 8.0%. Under this condition,the deproteinated rate was up to 85.1%,and the rate of polysaccharide loss rate was 16.3%. Conclusion: The papain-TCA may become a great candidate for Euryale petioles and pedicel polysaccharides purification.
