摘 要:目的:探索从提取甘蔗髓木糖母液中L-阿拉伯糖和D-木糖有效方法。方法:以PCR-642Ca2+树脂为固定相、纯水为流动相,使用连续色谱分离移动床对关键组分L-阿拉伯糖和D-木糖进行分离,探讨了Ⅰ区、Ⅱ区、Ⅲ区、Ⅳ区流量,进料流速,进料浓度等操作条件对连续移动床色谱分离效果的影响,对分离工艺条件进行了优化。结果:最佳工艺参数为进料浓度46.5%,进料流速9m L/min,Ⅰ区、Ⅱ区、Ⅲ区、Ⅳ区的流速分别为67.5、57.5、69、59 m L/min,L-阿拉伯糖出口流速14 m L/min,木糖出口流速12 m L/min。在此条件下,L-阿拉伯糖纯度达到90.01%,收率达到为91.52%;D-木糖纯度达到93.05%,收率达到90.88%。结论:为从甘蔗髓木糖母液等富含D-木糖、L-阿拉伯糖的料液,生产L-阿拉伯糖和D-木糖的高效、环保、成本低、生产工艺提供了参考。Objective: The aim was to establish high efficiency method of extracting L-arabinose and D-xylose from the xylose crystalline mother liquid of sugarcane leaves. Method: With resin of PCR-642 Ca2+ as the padding and deionized water as the mobile phase, continuous chromatographic separator bed was used to separate the key components of L-arabinose and D-xylose. The influences of operating conditions(flow rate in section Ⅰ、Ⅱ、Ⅲ、Ⅳ,feed flow rate, feed concentration) on the f continuous chromatographic separator bed separation effects were studied here. Result: The optimized parameters were :46.5% as feed concentration,9 m L/min as feed rate,67.5 m L/min as flow rate in section Ⅰ, 57.5 m L/min as flow rate in section Ⅱ, 69 m L/min as flow rate in section Ⅲ, 59 m L/min as flow rate in section Ⅳ,14 m L/min as L-arabinose rate,12 m L/min as D-xylose rate. Under these conditons, the purity of purified L-arabinose was 90.01% and the recovery was 91.52%,the purity of purified D-xylose was 93.05% and the recovery was 90.88%. Conclusion: This paper provide a reference in search of the xylose crystalline mother liquid from extraction of Bagasse pith enriched in D-xylose liquid and L-arabinose liquid, to produce L-arabinose and D-xylose in efficiency, environmental protection, low cost and easy to realize production process.
