摘 要:目的探讨高温常压纯水法与p H9.0的乙二胺四乙酸(Ethylene Diamine Tetraacetic Acid,EDTA)高温高压法对FISH检测乳腺癌HER2基因扩增表达结果的影响。方法选取皖南医学院弋矶山医院病理科2013年7月至2017年6月检测的40例浸润性乳腺癌标本,用高温常压纯水法(组一)与p H9.0的EDTA高温高压法(组二)进行FISH检测乳腺癌HER2基因扩增情况,并观察两组结果差异。结果荧光显微镜下观察,组一探针定位准确,红绿信号强度适中,亮度较强,背景清晰,视野干净易计数;组二探针信号定位不佳,锐利度明显下降,视野不干净。组一基因扩增阳性率显著高于组二(P<0.05),差异有统计学意义。结论高温常压纯水法在FISH检测乳腺癌HER2基因中可得到更准确且稳定的结果。Objective To explore the method of high temperature and atmospheric water and p H9.0 ethylenediamine Tetraacetic Acid(Ethylene Diamine Tetraacetic Acid, EDTA) high temperature and high pressure method for detecting the FISH have influence on the result of the expression of HER2 breast cancer gene amplification. Methods Select the Pathology Department of Wangnan Medical College Yijishan Hospital from July 2013 to June 2017 the detection of 40 cases of invasive breast cancer specimens, using the hightemperature atmospheric pressure pure water method(Group 1) and the EDTA high-temperature high-pressure method at p H 9.0(Group 2) to perform FISH detection of HER2 gene amplification in breast cancer, observe the difference between the two groups. Results The fluorescent microscope, group 1 probe location accurate, red and green signal intensity, moderate intensity is stronger, the background is clear, clean easy to count view; Group 2 probe signal poor positioning, sharpness decreases obviously, the vision not clean. A gene amplification positive rate was significantly higher than group 2(P〈0.05), the difference was statistically significant. Conclusion The method of high temperature and atmospheric pressure water in the FISH to detect breast cancer HER2 gene can obtain more accurate and stable results.
