摘 要:本研究用不同浓度的甲醇和乙醇法及蒸馏水法超声提取杜仲叶的活性成分,结果表明,杜仲叶总黄酮用甲醇提取效果较好,且在浓度为60%效果最佳,提取物总黄酮含量为207.006 mg QuE/g DM;杜仲叶总酚用乙醇提取效果较好,其浓度为60%最佳,提取物总酚含量为38.439 mg GAE/g DM。杜仲叶的60%甲醇提取物具有最佳的DPPH·清除能力和最佳的ABTS+·清除能力,其IC50值分别为72.86μg DM/mL和169.55μg DM/mL。50%乙醇提取物具有最佳的α-葡萄糖苷酶活性抑制能力,其IC50值为94.98μg DM/m L。相关性分析表明,杜仲叶总黄酮含量与DPPH·清除能力,ABTS+·清除能力和α-葡萄糖苷酶活性抑制能力之间的相关系数分别为-0.984,-0.891和-0.873,具有显著相关性。本研究通过筛选提取杜仲叶的活性成分的方法,为杜仲叶提取物的抗氧化性和降血糖能力的进一步研究提供了科学依据。The active components ofEucommia ulmoides leaves were extracted by ultrasonic with different conce- ntrations of methanol, ethanol and distilled water in this paper. The result showed that the total flavonoid of Eucommia ulmoides leaves was extracted better with methanol, and the best effect was in the concentration of 60%. The total flavonoids content of the extraction was 207.006 mg QuE/g DM. Ethanol was preferable for the extraction of the total phenol in Eucommia ulmoides leaves, and the best effect was in the concentration of 60%. The total phenol content of the extraction was 38.439 mg GAE/g DM. The extract of Eucommia ulmoides leaves with 60% methanol had the best DPPH·scavenging and ABTS+-scavenging ability, and the IC50 values were 72.86μg DM/mL and 169.55μg DM/mL, respectively. The 50% ethanol extract had the best inhibition ability of alpha glucosidase activity, and its IC50 value was 94.98 g DM/mL. Correlation analysis indicated that total flavonoid content of Eucommia ulmoides leaves had significant correlation with DPPH-scavenging capacity, ABTS+· clearance ability and inhibition ability of alpha glucosidase activity, and the correlation coefficients were -0.984, -0.891 and -0.873, respectively. This study provided a scientific basis for further research on the antioxida- nt activity and hypoglycemic ability of Eucommia ulmoides leaves by screening the extraction methods of the active components from leaves ofEucommia ulmoides.
