摘  要:目的探讨慢病毒介导的moesin基因沉默对U251细胞增殖与侵袭的影响。方法构建moesin基因沉默shRNA慢病毒载体并鉴定,将慢病毒表达载体稳定转染人胶质瘤细胞U251后,real time PCR方法与Western blot方法检测U251细胞moesin mRNA与蛋白的表达, MTT方法检测U251细胞增殖情况,Transwell实验检测细胞侵袭能力的改变。结果转染moesin基因沉默shRNA慢病毒载体后,人胶质瘤细胞U251 moesin mRNA和蛋白表达水平均明显降低,结果表明成功建立了稳定沉默moesin基因的U251细胞株。与空白对照组比较,在抑制moesin表达后,U251细胞增殖和侵袭能力也明显下降。结论成功建立了稳定沉默moesin基因的U251细胞株,moesin可能参与U251细胞增殖和侵袭的发病过程。Objective To investigate the effect of lentivirus mediated moesin gene silencing on the proliferation and invasion of U251 cells. Methods To construct silencing moesin gene shRNA lentiviral vector, and the expression level of moesin mRNA and protein in U251 cells was detected by real time PCR and Western blot. The proliferation of U251 cells was observed by MTT, and invasion ability of U251 cells was observed by Transwell assay. Results The expression levels of moesin mRNA and protein in U251 cells was decreased significantly after the lentiviral vector was transfected, which suggested that U251 cell line with stable silencing of moesin gene was successfully established. Compared with the blank control group, the proliferation and invasion ability of U251 cells was decreased significantly after the expression of moesin was inhibited. Conclusion U251 cell line with stable silencing of moesin gene was successfully established, moesin gene might be involved in the pathogenesis of U251 cells.
