摘 要:目的探讨白藜芦醇对哮喘幼鼠肺组织toll样受体4(TLR4)与β干扰素TIR结构域衔接蛋白(TRIF)表达的影响。方法 45只BALB/c幼鼠随机分为对照组、哮喘组和白藜芦醇组。采用卵清蛋白(OVA)刺激,建立幼鼠哮喘模型。HE染色观察各组幼鼠肺组织病理学改变,测定各组幼鼠支气管肺泡灌洗液(BALF)中白细胞总数及嗜酸粒细胞(EOS)数,Western blot方法检测各组幼鼠肺组织TLR4与TRIF的蛋白表达变化,real time PCR方法检测各组幼鼠肺组织TLR4 mRNA与TRIF mRNA的表达水平。结果与对照组相比,哮喘组幼鼠肺组织可见明显的炎性细胞浸润,气道管壁明显增厚,BALF中细胞总数及嗜酸粒细胞计数显著升高,P<0.01;与哮喘组相比,白藜芦醇组幼鼠肺内炎性细胞浸润明显减轻,BALF中细胞总数及嗜酸粒细胞计数显著降低,P<0.01。与对照组相比,哮喘组幼鼠肺组织TLR4与TRIF的蛋白与mRNA表达水平显著升高,P<0.01;与哮喘组相比,白藜芦醇组幼鼠肺组织TLR4与TRIF的蛋白与mRNA表达水平显著降低,P<0.01。结论白藜芦醇能够抑制哮喘幼鼠肺组织TLR4与TRIF的表达。Objective To investigate the effect of resveratrol on the expression of toll-like receptor 4(TLR4) and TIR-domain containing adaptor inducing interferon-β(TRIF) in lung tissue of asthmatic mice. Methods 45 BALB/c mice were randomly divided into control group, asthma group and resveratrol group. A mouse asthma model was established by ovalbumin(OVA) stimulation. The pathological changes of lung tissue in each group were observed by HE staining, and the number of total leukocytes and eosinophils(EOS)in bronchoalveolar lavage fluid(BALF)was counted. The protein expression of TLR4 and TRIF in lung tissues of all mice was detected by Western blot method. Real time PCR method was used to detect the level of TLR4 mRNA and TRIF mRNA of lung tissue in each group. Results Compared with the control group, the lung tissues of mice in asthmatic group showed obvious inflammatory cell infiltration, the airway tube wall thickened obviously, and the total number of total leukocytes and eosinophils in the BALF was increased significantly, P〈0.01. Compared with the asthma group, the infiltration of inflammatory cells in the lung of mice in resveratrol group was significantly reduced, the total number of total leukocytes and the eosinophil in the BALF were decreased significantly, P〈0.01. Compared with the control group, the protein and mRNA expression level of TLR4 and TRIF in the lung tissue of mice in the asthmatic group was increased significantly, P〈0.01. Compared with the asthma group, the protein and mRNA expression level of TLR4 and TRIF in the lung tissue of mice in the resveratrol group was decreased significantly, P〈0.01. Conclusion Resveratrol could inhibit the expression of TLR4 and TRIF in lung tissue of asthmatic mice.
