摘 要:探究超声辅助酶法制备米糠黄酮的最佳工艺,以及制备的米糠黄酮对人肺癌细胞(A549)生长的抑制作用。通过单因素和响应面实验,研究超声辅助酶法制备米糠黄酮的最佳条件。研究结果显示,提取米糠黄酮的最佳条件为:料液比1:19 g/mL、加酶量125 U/g、乙醇体积分数60%、超声时间19 min,在此条件下得率为0. 52%;通过形态学观察、MTT法检测细胞存活率及Hoechst33258检测凋亡作用等细胞实验显示,米糠黄酮对A549细胞生长有一定的抑制作用。实验结果表明,超声辅助酶法提取米糠黄酮是一种有效的提取方法,制备的米糠黄酮在细胞水平显示了潜在的抗癌活性。This paper was to investigate the best preparation technology of rice bran flavonoids by ultrasonic assisted with enzymatic method,and the inhibitory effect of rice bran flavonoids on the growth of human lung cancer cells (A549). The optimal conditions for the preparation of rice bran flavonoids by enzymatic method assisted with ultrasonic were studied by single factor and response surface test. The optimal extraction condition of rice bran flavonoids was as follow: solid-liquid ratio 1∶ 19 g/ml,enzyme quantity 125 U/g,ethanol volume fraction 60%,ultrasonic time 19 min; the extraction yield was 0. 52%. The inhibitory effect of rice bran flavonoids on A549 cells was detected by morphological observation,MTT assay and Hoechst33258 to evaluate the anticancer activity. The results of cell experiments showed that the flavonoids from rice bran could inhibit the growth of A549 cells. The present study revealed that the extraction of rice bran flavonoids by ultrasonic assisted with enzymatic method was a kind of effective extraction method,the rice bran flavonoids prepared by this method exhibited a potential anticancer activity in cellular level.
