摘 要:根据金针菇(Flammulina velutipes)基因组及转录组数据,筛选得到1个β-1,6-葡聚糖合酶(β-1,6-glucan synthase)编码基因,命名为fv-gs6.该基因全长1 439 bp,含有9个外显子,8个内含子,编码339个氨基酸.多序列比对及系统进化分析结果表明:fv-gs6编码蛋白(Fv-GS6)具有β-1,6-葡聚糖合酶的保守motif,且与担子菌的β-1,6-葡聚糖合酶聚在同一个分支.生物信息学分析表明:该蛋白具备信号肽、亚细胞定位于细胞外,属于分泌蛋白;推测Fv-GS6蛋白可能直接在细胞外催化β-1,6-葡聚糖的合成.通过qRT-PCR方法检测该基因在子实体不同发育时期的表达量,结果显示:fv-gs6在菌柄上高表达,且在伸长期菌柄表达量最高;其中,菌柄的伸长区段表达量是不伸长区段的5.42倍.根据β-1,6-葡聚糖合酶在真菌细胞壁合成中的功能,推测fv-gs6的高表达可能促进细胞壁合成,参与金针菇菌柄的伸长过程.Based on genome and transcriptome data of Flammulina velutipes, a β-1, 6-glucan synthase coding gene named fv-gs6was studied. fv-gs6 contained 1 439 bp, encoding 339 amino acids. It contained 9 exons and 8 introns. Multi-sequence alignment andphylogenetic analysis showed that protein Fv-GS6 that fv-gs6 encoded had a conserved motif of β-1, 6-glucan synthase and was clus-tered with β-1, 6-glucan synthase of Basidiomycetes. Bioinformatics analysis showed that protein Fv-GS6 contained signal peptide,and the subcellular localization of it was extracellular, suggesting that Fv-GS6 could directly catalyze the synthesis of β-1, 6-glucan.Then relative expression level of fv-gs6 in different growth and development stages of fruiting body was analyzed by qRT-PCR. Theresults showed that the relative expression level of fv-gs6 was the highest in the stipe, especially in the elongation stage. Comparedwith non-elongated region, the expression level of fv-gs6 in elongated region of stipe was about 5.42 times. According to the functionof β-1, 6-glucan synthase in fungal cell wall synthesis, it is speculated that the high expression of fv-gs6 may promote cell wall syn-thesis and participate in the stipe elongation of F.velutipes.
