摘 要:为提高罗布麻生物脱胶效果,采用菌株复合培养发酵方式进行生物脱胶。以响应面法为工具对发酵条件进行优化,以麻皮生物脱胶后的失重率为脱胶效果的评价指标,通过单因素试验,筛选出摇床转速、发酵初始pH值、浴比、MgSO_4·7H_2O质量浓度4个显著影响因素。以Design-Expert法得出最佳发酵条件:摇床转速77 r/min、初始pH值8、浴比1:40、MgSO_4·7H_2O质量浓度0.069g/L、发酵温度40℃、菌液接种量30%、脱胶时间4 d,槐糖脂质量浓度0.15 g/L。在最优条件下,罗布麻韧皮经混菌发酵脱胶后失重率为29.1%,比优化前失重率提高了1.6倍。The mixed bacteria fermentation method of degumming is adopted to improve the apocynum bast biodegumming effect. The fermentation conditions are optimized by response surface method, and the weight loss rate of bast after degumming is taken as the evaluation index. Through single factor ex- periment, 4 significant influencing factors are screened out: shaker speed, initial pH, bath ratio, additive amount of MgSO4-7H20. The optimum fermentation conditions are obtained by response surface meth- od. The optimal submerged culture conditions are: shaker speed 77 r/min, initial pH value 8, liquor ratio 1:40, MgSO4.7H20 0.069 g/L, fermentation temperature 40℃, inoculum size of bacteria 30%, fermenta- tion time 4 day and sophorolipid concentration 0.15 g/L. Under the optimal condition, the weight loss rate of apocynum bast after biodegumming is 29.1%, which is close to the predicted value of 29.89%, and the weiClht loss rate increased by 1.6 times compared with that before optimization.
