摘  要:分析比较日本刺参多糖和大西洋海参多糖的化学组成和对血管内皮细胞的保护作用。采用酶解法和乙醇沉淀从2种海参体壁中提取酸性粘多糖,以总糖、蛋白、硫酸根、糖醛酸、氨基糖和中性单糖含量为指标,进行化学组成分析比较,并在Q-Sepharose FF阴离子交换树脂色谱柱上分离,绘得洗脱曲线。采用氧化低密度脂蛋白(ox-LDL)造成体外培养的人脐静脉内皮细胞株ECV304细胞损伤模型,用MTT法检测2种酸性粘多糖对血管内皮细胞的增殖活性。结果表明,2种酸性粘多糖在总糖含量和葡萄糖含量上差异最为明显,日本刺参多糖为62.11%和6.19%,而大西洋海参多糖仅为41.69%和0.42%。日本刺参多糖在Q-Sepharose FF阴离子交换树脂色谱柱上被完全洗脱的盐浓度为3.8 mol.L-1,而大西洋海参多糖的为2.6 mo.lL-1。100μg.mL-1日本刺参多糖和大西洋海参多糖对损伤ECV304细胞的增殖率分别为66.62%和58.63%,日本刺参多糖的细胞保护作用更优。This study aims to analyze the chemical composition differences of the polysaccharides extracted from sea cucumbers—Apostichopus japonicus and Holothuria mexicana—and their protective effects on vascular endothelial cells.The polysaccharides were extracted by papain,precipitated by ethanol,analyzed with six chemical indexes,including the contents of total sugars,protein,sulfate,uronic acid,amino sugars and neutral monosaccharide,and eluted via Q-Sepharose FF anion-exchange chromatography,which presented the elution curves.A model of endothelial cell oxidative damage induced by ox-LDL was established to test the influence of the two polysaccharides on the proliferation activity of endothelial cell oxidative damage using MTT assay.The results indicate that the two polysaccharides show great differences in the indexes of contents of total sugars and glucose,which are 62.11% and 6.19% respectively for the polysaccharide from Apostichopus japonicus(P-AJ),and 41.69% and 0.42% respectively for the polysaccharide from Holothuria mexicana(P-HM).The ultimate elution concentration of P-AJ on Q-Sepharose FF is 3.8 mol·L-1;while that of P-HM is 2.6 mol·L-1.The proliferation ratio to the injured vascular endothelial cell of the two polysaccharides at the concentration of 100 μg·mL-1 are 66.62% and 58.63%,respectively indicating that P-AJ works better.
