摘  要:目的:探讨体外c-myc反义寡核苷酸(AODN)逆转人卵巢癌顺铂(DDP)耐药的可行性。方法:以卵巢癌DDP耐药细胞株COC1/DDP为研究对象,实验组通过脂质体为载体,分别将c-mycAODN、c-myc无义寡核苷酸(SODN)转染到OC1/DDP细胞内,阴性对照组细胞以同体积的培养基转染。采用计数法检测细胞增殖变化;四甲基偶氮唑蓝(MTT)法检测DDP对细胞增殖的抑制率;RT-PCR、免疫组织化学技术分别对体外培养细胞c-mycmRNA及蛋白的表达进行检测,研究c-mycAODN逆转癌细胞耐药的作用。结果:实验组转染AODN的COC1/DDP细胞增殖被抑制;实验组转染AODN的卵巢癌细胞对DDP的敏感性提高,细胞生长抑制率高,与SODN组及阴性对照组比较,差异均有统计学意义(P值均<0.05);AODN转染组COC1/DDP细胞的c-mycmRNA及蛋白表达均明显降低,与相同浓度SODN转染组、阴性对照组COC1/DDP细胞比较,差异均有统计学意义(P值均<0.05)。结论:c-mycAODN能有效逆转卵巢癌细胞DDP耐药性;c-myc反义寡核苷酸可望成为一种有效的卵巢癌治疗方法。Objective:To evaluate the in uence of c-myc antisense oligodeoxynucleotides(AODN)on reversing cisplatin(DDP) resistance in ovarian cancer.Methods:In the experiment groups,oligodeoxynueleotides were transfected into cisplatin resistant ovarian carcinoma cell line COC1/DDP cells by lipofectamine.Cell proliferation was determined by counting the number of the cells continuously for six days.Then they were treated with cisplatin and the inhibitory rates of cells were detected by MTT.The effects of the reversion of carcinoma cells'drug resistance and the reduction of their malignancy and growth capability were assessed using RT-PCR,immunohistochemistry techniques.The cells treated with culture media were used as the negative contro1.Results:The proliferation of AODN transfected cells was inhibited significantly compared with that in SODN and negative groups.The AODN transfected cells were sensitive to DDP again,and the inhibitory rate was higher than that of the negative control(P<0.05).The expressions of c-myc mRNA and protein in AODN transfected cells decreased were compared with the expressions in the negative and SODN groups,the difference were significant(P<0.05).Conclusion:AODN can reverse the resistance to cisplatin in ovarian cancer;So it may serve as a new treatment method for ovarian cancer.
