摘  要:目的 :研究端粒酶反义寡核苷酸对鼻咽癌细胞生长分化的影响。方法 :脂质体介导反义寡核苷酸转染鼻咽癌CNE1和CNE2Z细胞株 ,用细胞体外增殖实验、免疫组织化学S P法 ,观察细胞生长分化的变化情况。结果 :针对端粒酶RNA模板区的反义寡核苷酸可抑制CNE1和CNE2Z细胞的增殖并呈剂量和时间依赖性 ,反义寡核苷酸组细胞角蛋白表达高于其它处理组及空白对照组 (P <0 0 1) ,细胞形态趋向分化成熟。结论 :端粒酶反义寡核苷酸可抑制鼻咽癌CNE1和CNE2Z细胞的生长 ,并促进细胞分化。Purpose To study the effects of telomerase antisense oligodeoxynucleotides (ASODN) on CNE1 and CNE2Z cell lines of nasopharyngeal carcinoma cells (NPC). Methods After transfecting telomerase ASODN into NPC CNE1 and CNE2Z cell lines by lipoffectin, the proliferation, morphological changes and keratin expression of the cells were studied. Results ASODN targeted at telomerase RNA template sequence acted as a specific growth inhibitor to CNE1 and CNE2Z cell lines, and presented in a dose and time dependent manner. At the same time, the morphology of the cells also changed obviously, and the keratin expression of ASODN group was significantly higher than that of the other treatment groups and control group ( P <0 01). Conclusion Telomerase ASODN is able to inhibit the growth of CNE1 and CNE2Z cell lines of NPC, and induce the cell differentiation into morphologically mature.
