摘 要:目的采用气泡上升法测定蚓激酶效价。方法在37℃环境下,在试管中依次加入纤维蛋白原、蚓激酶,以及凝血酶和纤维蛋白溶酶原的混合溶液,迅速混匀计时,待气泡上升至整个体系1/2时停止计时。以标准品溶液浓度对数为横坐标,时间对数为纵坐标进行线性回归,通过线性回归方程计算样品蚓激酶的效价。结果线性回归方程为y=-1.1513x+4.3136,R2=0.9969,精密度及重复性RSD值分别0.73%及6.01%。结论气泡上升法能够测定蚓激酶的双重活性,更好地反应出蚓激酶的效价特点。Objective To determine the activity of lumbrokinase by the bubble rise method.Methods In the37℃environment,added fibrinogen,lumbrokinase,thrombin and plasminogen were added in the test tube in turn,quickly mixed and timed,stopped timing until the bubble rose to the half of the whole system.The logarithm of the concentration of the standard solution was the abscissa,and the logarithm of the time was the ordinate.The activity of the lumbrokinase was calculated by the linear regression equation.Results The linear regression equation was y=-1.1513x+4.3136,R2=0.9969.The accuracy and repeatability were0.73%and6.01%.Conclusion The bubble rise method could determine the double activities of lumbrokinase,better response the characteristics of lumbrokinase.
