摘  要:为探讨SREBP1基因多态性及其与肌内脂肪酸的相关性,选取30月龄,健康无病,体重相近的延边黄牛29头,屠宰后采集肉样与血样,进行DNA提取与脂肪酸含量测定,利用PCR方法检测固醇调节元件结合蛋白(Slerol regulatory element binding factor 1,SREBP1)基因第5内含子的多态性,并与肌内脂肪沉积性状进行相关性分析。结果表明:在延边黄牛SREBP1基因第5内含子发现84bp插入缺失突变,且与肌内脂肪酸组成显著相关。AB型个体的月桂酸(C12∶0)、棕榈油酸(C16∶1)、硬脂酸(C18∶0)、亚油酸(C18∶2)含量显著高于AA型个体(P<0.05)。说明延边黄牛SREBP基因第5内含子存在84bp插入缺失突变,且这一位点的多态性影响肌肉脂肪酸组成。The aim of this study was to investigate the polymorphism of SREBP1gene and its correlation with intramuscular fatty acids.Twenty nine healthy30month old Yanbian yellow cattles with similar weight were selected.The meat samples and blood samples were collected after slaughter.The polymorphism of the fifth intron of the Slerol regulatory element binding factor1(SREBP1)gene was detected by polymerase chain reaction(PCR),and the correlation analysis was performed with the intramuscular fat deposition traits.The results showed that the deletion of84bp was found in the5th intron of SREBP1gene in Yanbian yellow cattle and it was significantly correlated with the composition of intramuscular fatty acid(C12∶0),palmitoleic acid(C16∶1),stearic acid(C18∶0)and linoleic acid(C18∶2)of AB type were significantly higher than those of AA type(P<0.05),suggesting that the5intron of the SREBP gene of Yanbian yellow cattle had84bp insertion deletion mutation,and the polymorphism of this site affected the composition of muscle fatty acid.
