摘  要:目的探索水提醇沉法提取红花多糖的最佳提取工艺,测定塔城、伊犁、和田3个不同产地红花中多糖的含量,并考察其体外抗氧化活性。方法在单因素实验的基础上,采用Box-behnken中心组合实验原理,以料液比、提取温度、提取时间为自变量,多糖提取率为响应值,设计3个因素3个水平实验,进行红花中多糖最佳提取工艺的优化。采用蒽酮-硫酸比色法测定红花中多糖的含量。以伊犁红花为研究对象,采用DPPH·、·OH及铜离子还原实验3种方法评价红花多糖的体外抗氧化活性。结果各因素对红花多糖提取率的影响次序依次为:提取温度>提取时间>料液比,最佳提取工艺为:料液比1∶51(g/mL),提取温度94℃,提取时间1.8h。测得塔城、伊犁、和田3个地区红花多糖的含量依次为(32.7±6.2)、(34.6±8.4)、(28.9±7.6)mg/g。红花多糖对DPPH·和·OH有较好的清除能力,对铜离子有较强的还原能力,且呈一定的量效关系。结论该提取方法简便易行,结果准确可靠,适用于红花多糖的提取,红花多糖具有较强的抗氧化活性,有望被开发成为天然的抗氧化剂。ObjectiveTo optimizethe water extract and ethanol precipitationtechnology for extracting safflower polysaccharide,and determine the polysaccharide contents of safflower in Tacheng,Ili and Khotan.Theantioxidative activity of safflower polysaccharide was also evaluated in vitro.MethodsOn the basis of single factor experiment,the optimal extraction technology was obtained by Box-behnken design and response surface methodology with three factors and three levels.The independent variables were solid-liquid ratio,extraction temperature,and extraction time.The response value was extraction rate of polysaccharide.Anthrone-sulphuric acid colorimetricmethod was used to determine the polysaccharide contents of safflower.Theantioxidative activity of polysaccharide,taken from safflower in Ili,was also evaluated in vitro by DPPH·,·OH and copper ion reduction method.ResultsThe results showed that the influence of various factors on the extraction yield were as follow:extraction temperature>extraction time>solid-liquid ratio.The optimum extracting conditions are solid-liquid ratio 1∶51(g/mL),extraction temperature at 94℃,extraction time is 1.8 h.Under this conditions,thecontents of safflower polysaccharide in Tacheng,Ili and Khotan were(32.7±6.2)mg/g,(34.6±8.4)mg/g,(28.9±7.6)mg/g,respectively.The results of antioxidation experiment showed that safflower polysaccharide has good ability to clear DPPH·and·OH,reduce copper ion in a dose-effect relationship.ConclusionThe established extraction method is simple and accurate,which is suitable for the extraction of safflower polysaccharide.Because of its strong antioxidative activity,s afflower polysaccharide could be explored as potential natural antioxidant.
