摘  要:建立高效液相色谱-蒸发光散射检测器法(HPLC-ELSD)测定酵母菌株发酵液中多元醇含量的方法。采用Agilent Hi-plex Ca色谱柱(7.7 mm×300 mm,8μm),以纯水为流动相,流速0.5 m L/min,蒸发光散射检测器(ELSD)为检测器;柱温为70℃,进样量为4μL。结果表明,5种多元醇在0.8~6.0 mg/m L质量浓度范围内与峰面积呈良好的线性关系(相关系数R均≥0.996);精密度实验结果相对标准偏差(RSD)均<5%,平均回收率为96.73%~100.46%,表明该方法灵敏度高,重现性良好,准确度高,可用于发酵液中5种多元醇的检测。以酱香型白酒酿造过程中大曲和酒醅分离的酵母为研究对象,运用该方法在酵母发酵液中检测到5种多元醇。The method for determination of polyhydric alcohols content in yeast fermentation broth was established by HPLC-ELSD.The samples were separated on Agilent Hi-plexCa chromatographic column(7.7 mmx300 mm,8 pm),with pure water as mobile phase at a flow rate of 0.5 ml/min.The evaporative light-scattering detector(ELSD)was used as detector.Column temperature was 70^and the injection volume was 4 pl.The results showed that 5 polyhydric alcohols had a good linear relationship with peak area in the range of 0.8-6.0 mg/ml(R^0.996).The relative standard deviation(RSD)of precision experiment results was less than 5%,the recovery rate was 96.73%-100.46%.The result indicated that the method showed high sensitivity,good reproducibility and high accuracy,and it could be used for the detection of 5 kinds of polyhydric alcohols in fermentation broth.Using the yeast separated^om Daqu and fermented grains during the brewing process of Moutai-flavor Baijiu as the research object,5 polyhydric alcohols in yeast fermentation broth could be detected by the method.
