摘  要:目的探讨牛磺酸(Tau)对人肺癌细胞株A549细胞凋亡的作用及其初步机制。方法将A549细胞分为6组:Control组(正常对照组,未给予任何药物处理)和Tau 1组(给予Tau 10 mmol·L^(-1))、Tau 2组(给予Tau20mmol·L^(-1))、Tau 3组(给予Tau 40 mmol·L^(-1))、Tau 4组(给予Tau 80 mmol·L^(-1))、Tau 5组(给予Tau160mmol·L^(-1));阳性对照组DDP 4mg·L^(-1),作用时间分别为24、48、72h。应用MTT比色法检测人肺癌细胞株A549的细胞增殖,流式细胞术检测人肺癌细胞株A549的细胞凋亡率,分光光度法检测Caspase-9、Caspase-3的活性,免疫印迹法检测p53正向细胞凋亡调控因子(PUMA)的水平及Bax蛋白、Bcl-2蛋白的表达水平。结果Tau能够以时间剂量依赖方式来抑制A549细胞增殖,并显著增加A549细胞凋亡率。Tau可以明显增加A549细胞内Caspase-9、Caspase-3的活性,以及诱导A549细胞内PUMA、Bax蛋白表达上调,Bcl-2蛋白表达下调。Tau对人胚肾细胞293T的细胞活性无明显影响。结论 Tau对人肺癌细胞株A549有增殖抑制和凋亡诱导的作用,其机制可能是Tau通过上调PUMA、Bax表达及下调Bcl-2表达,进而增加Caspase-9、Caspase-3的活性来诱导A549细胞发生凋亡。Objective To investigate the effect of taurine(Tau)on the apoptosis of human lung cancer A549 cells and its mechanism of action.Methods A549 cells were divided into 6 groups:control group(normal control group,no treatment),Tau 1 group(treatment with 10 mmol·L-1 Tau),Tau 2 group(treatment with 20 mmol·L-1 Tau),Tau 3 group(treatment with 40 mmol·L-1 Tau),Tau 4 group(treatment with 80 mmol·L-1 Tau),and Tau 5 group(treatment with 160 mmol·L-1 Tau).Cells were cultured with 4 mg·L-1 DDP as the positive control group.All groups were observed for 24,48 and 72 hours.The proliferation and apoptosis of cells were detected by MTT colorimetric assay and flow cytometry,respectively.The activities of caspase-9 and caspase-3 were measured by spectrophotometry.The expression of p53 up-regulated modulator of apoptosis(PUMA),Bax and Bcl-2 was determined by Western blot.Results The Tau inhibited the proliferation of A549 cells in a dose-dependent manner and increased the apoptosis rate of A549 cells.Furthermore,Tau elevated the activities of caspase-9 and caspase-3,up-reguated the expression of PUMA and Bax,and down-regulated the expression of Bcl-2 in A549 cells.Tau had no significant effect on the viability of human embryonic kidney 293T cells.Conclusion Tau can inhibit the proliferation and induce the apoptosis in human lung cancer A549 cells through up-regulating PUMA and Bax expression and down-regulating Bcl-2 expression to increase caspase-9 and caspase-3 activities.
