摘 要:为了改善高胆固醇血症,对红米麸皮中花色苷进行优化提取,通过筛选大孔树脂获得花色苷富集工艺条件,并研究红米花色苷对胆固醇消化吸收的影响.实验结果显示,红米花色苷提取最优条件:乙醇体积分数75%、pH值0.5、温度40℃、料液比1∶10、提取时间2.5 h、提取次数3次.大孔树脂HPD100对花色苷具有较高的吸附和解吸率,最佳富集工艺:上样体积140 mL、温度25℃、吸附1.5 h、洗脱液乙醇体积分数80%、流速1.0 mL/min、洗脱体积20 mL.红米花色苷主要成分为矢车菊素-3-葡萄糖苷(Cy-3-G),经大孔树脂富集处理,Cy-3-G含量达36.48%.红米花色苷对胰脂肪酶具有抑制作用,Cy-3-G含量越高,抑制作用越强.红米水提取物、乙醇提取物、大孔树脂富集物对胰脂肪酶活力抑制的IC50分别为61.13 mg/L、30.11 mg/L、18.91 mg/L.红米花色苷具有降低胆固醇微团溶解度的作用,Cy-3-G含量越高,降低胆固醇溶解作用越强.研究表明,红米及其花色苷是通过抑制胰脂肪酶活性和降低胆固醇微团溶解度起到调节血脂的作用.In order to improve hypercholesteremia syndromes,this study investigated the inhibition of cholesterol absorption in vitro by red rice anthocyanins.The purification conditions of red rice anthocyanins were established by selecting macroporous resin.The results showed that the optimal extraction parameters contain ethanol concentrations 75%,pH value 0.5,temperatures 40°C,solvent ratios 1∶10,extraction time 2.5 h,extraction cycles 3.The adsorption and desorption rates of red rice anthocyanins were high by using macroporous resin HPD100.The optimal extraction processing parameters contain load sample volumes 140 mL,temperatures 25°C,adsorption time 1.5 h,eluent ethanol concentrations 80%,current speeds 1.0 mL/min,eluent volumes 20 mL.The main content of red rice anthocyanins was Cyanidin-3-glucoside(Cy-3-G).The purity of Cy-3-G in a fraction of macroporous resin was 36.48%.Red rice anthocyanins reduced cholesterol absorption by inhibiting pancreatic lipase and decreasing the micellar cholesterol solubility.The inhibitory activity was positively associated with Cy-3-G content of the extracts.The IC50 values of red rice extracts which were aqueous,ethanol and a fraction of macroporous resin against pancreatic lipase were 61.13 mg/L,30.11 mg/L,and 18.91 mg/L,respectively.All these data indicated that red rice anthocyanins might be applied for the prevention of hypercholesterolaemia by inhibiting pancreatic lipase and decreasing the micellar cholesterol solubility.
