摘  要:以扳倒井芝麻香型白酒高温大曲中分离的菌株CICC 33077为研究对象,采用多相分类学技术确定该菌株的分类学地位;以生物量(OD_(600nm))为响应值,并通过Plackett-Burman试验、最陡爬坡试验以及Box-Behnken试验对其培养基组分进行优化。研究结果表明,菌株CICC 33077鉴定为扣囊复膜孢酵母(Saccharomycopsis fibuligera),最优培养基组分为:麦芽浸粉3.586%,大豆蛋白胨1.216%,蛋白胨1.470%,磷酸氢二钾1.148%;最适生长温度为30℃,最适生长pH值为6.0。菌株CICC 33077优化后的培养基中生长的生物量(OD_(600nm)值)能够达到(8.136±0.102),比优化前提高了170%。Strain CICC 33077 isolated from high-temperature Daqu of Bandaojing sesame-flavor Baijiu was identified by polyphasic taxonomy technique and the compositions of the medium were optimized using Plackett-Burman experiments,the steepest the steepest ascent experiments and Box-Behnken experiments with biomass(OD600 nm)as the response value.The results indicated that strain CICC 33077 was identified as Saccharomy-copsis fibuligera.The optimimum compositions of medium were as follows:malt extract powder 3.586%,soybean peptone 1.216%,peptone 1.470%,dipotassium hydrogen phosphate 1.148%.Meanwhile,the optimum growth temperature and pH value of strain CICC 33077 were 30℃and 6.0,respectively.The biomass(OD600600 nm)reached 8.136±0.102 when strain CICC 33077 was cultured in the optimum medium,which was 170%higher than that of before optimization.
