摘 要:用大孔树脂耦合硅胶柱层析法对小曲酒酿造废水中的高粱红色素进行分离纯化,通过单因素试验,确定了大孔树脂和硅胶层析柱的纯化条件。结果表明,一级纯化选择HPD600型大孔树脂,吸附容量为2BV,吸附液pH值为7,吸附速率为3BV/h,除杂水用量为5BV,洗脱剂为体积分数80%的乙醇,洗脱剂用量为2BV,洗脱速率为6BV/h;二级纯化用硅胶层析柱,流动相为石油醚∶乙酸乙酯=4∶1(V/V),目标收集液为2~3BV段的流动相收集液;经两级纯化后得到高粱红色素纯度达90%,废水中高粱红色素的回收率达67.2%。The separation of sorghum red pigment in the wastewater of Baijiu fermentation was optimized by the macroporous adsorption resin coupled silica gel column chromatography.Through the single factor experiments,the purification conditions of macroporous resin and silica gel column were determined.The results showed that the HPD600 macroporous resin was selected for primary purification,the adsorption capacity was 2 BV,the adsorption solution pH was 7,the adsorption rate was 3 BV/h,the volume of edulcoration water was 5 BV,the elution was ethanol with concentration 80%,the volume of eluent was 2 BV,the elution rate was 6 BV/h.The silica gel column was selected for secondary purification,the mobile phase was petroleum ether:ethyl acetate=4:1(V/V),the target collection fluid was 2-3 BV mobile phase collection fluid.After two levels of purification,the purity of the sorghum red pigment was 90%.The recovery rate of red sorghum pigment in wastewater was up to 67.2%.
