摘 要:本研究以绍兴黄酒熟麦曲为研究对象,采用丙酮法和Clean Up Kit试剂盒法来处理样品,发现Clean Up Kit试剂盒法能够有效去除盐离子、多糖等干扰物质,蛋白点数目较多且清晰,无拖尾现象。蛋白样品上样量为90μg时,分离效果较好。初步建立适用于绍兴黄酒熟麦曲的宏蛋白质组的样品制备方法,获得了较好的双向电泳图谱。In this study,Shaoxing koji was the research object,two different preparation methods of sample,acetone precipitation and Clean Up Kit were used to isolate the metaproteome.The results showed that Clean Up Kit can effectively remove salt ions,polysaccharides and other,the number of protein spots was more and clear.When the amount of protein was 90μg,the metapromics spots were effiectively separated.In conclusion,a suitable preparation method for metaproteome from Shaoxing rice wine koji was established,the two-dimensional electorphoresis maps with good quality were obtained.
