摘　　要：目的检测Smac/DIABLO在子宫内膜癌中的表达并探讨其发挥作用的可能机制。方法荧光定量PCR检测Smac/DIABLO在子宫内膜癌、非典型增生内膜和正常内膜中的不同表达,分析其与内膜癌临床病理分期的关系。在子宫内膜癌Ishikawa细胞中上调Smac/DIABLO表达,Western blot检测通路相关蛋白Caspase-3,7,9表达。结果 Smac/DIABLO在子宫内膜癌组表达量明显减少(P <0.01),其表达与FIGO分期、淋巴结转移及肌层浸润深度有关(P <0.05),与年龄、组织分化程度无关(P> 0.05)。过表达Smac/DIABLO后通路相关蛋白Caspase-3,7,9表达量明显增加(P <0.01,P <0.05)。结论 Smac/DIABLO表达与内膜癌FIGO分期、淋巴结转移及肌层浸润深度相关,推测其通过线粒体和死亡受体介导的两条凋亡通路发挥作用。Objective To detect the expression of Smac/DIABLO in endometrial carcinoma and explore its possible mechanisms.Methods qRT-PCR was used to detect the expression of Smac/DIABLO in endometrial carcinoma.The relationship between Smac/DIABLO and clinical pathological staging was analyzed.Smac/DIABLO was up-regulated in Ishikawa cells and Caspase-3,7,9 protein was detected by Western blot.Results The expression of Smac/DIABLO was decreased in endometrial carcinoma group(P<0.01).It was related to FIGO stage,lymph node metastasis,and myometrial infiltration depth(P<0.05).Caspase-3,7,9 protein was increased with the over-expression of Smac/DIABLO,(P<0.01,P<0.05).Conclusions Smac/DIABLO is associated with FIGO stage,lymph node metastasis,and myometrial infiltration depth of endometrial carcinoma.Smac/DIABLO plays a role through two apoptotic pathways mediated by mitochondria and death receptors.
