摘 要:该研究分别以植酸钙和卵磷脂为唯一磷源,采用解磷圈法和液体摇瓶培养法,从采自岷县麻子川乡曾使用过毒死蜱等其他有机磷农药的当归根系土壤中分离筛选有机磷降解菌,对其进行形态学、生理生化特性、分子生物学鉴定及生长特性征分析。结果表明,从当归根系土壤中分离纯化出一株能对有机磷(植酸钙)有降解能力的菌株,命名为w-3。经鉴定和生长特性测定表明,菌株w-3为枯草芽孢杆菌(Bacillus subtilis),其最适生长温度为35℃,最适初始pH值为8.0。菌株w-3具有良好的解磷能力,在多功能微生物菌肥研制中具有重要利用价值。Using phosphate-degrading circle method and liquid shake flask culture method,phytin and lecithin were used as the only phosphate source to screening organic phosphate degrading bacteria from soil sample collected from various locations in Minxian Mazichuan village that used some organophosphorus pesticides such as chlorpyrifos.The strain was identified by morphology,physicochemical characteristics and molecular biology identification and its growth characteristics were analyzed.The results showed that a strain named w-3 that an organic phosphorus(calcium phytate)-degrading was isolated and purified from the root system soil of Angelica sinensis.After identification and growth characteristics determination,strain w-3 was identified as Bacillus subtilis.The most suitable culture conditions of strain w-3 were temperature 35℃,initial pH 8.0.Consequently,the strain w-3 had good phosphorous degradation ability,thus it had high utilization value in developing multiple-effective microbial fertilizer.
