摘 要:对嗜酸乳杆菌进行ARTP多轮诱变处理,筛选出1株亚油酸异构酶高产菌株,研究了碳源、氮源、碳氮比、培养时间、培养温度、初始pH对其产酶的影响,并通过响应面实验对产酶条件进行了优化.结果表明,最佳碳源为2%的葡萄糖,最佳氮源为2%的牛肉膏,当碳氮比为1∶1.5,培养时间为36h,培养温度为36.4℃,初始pH为5.6时,诱变菌株酶活达到1 214.1U/mL,是原始菌株的2.84倍.A high productivity strain of linoleic acid isomerase was obtained by ARTP mutagenesis in Lactobacillus acidophilus.The effects of carbon,nitrogen,C/N,cultivation temperature,cultivation time and initial pH on synthesis of linoleic acid isomerase were studied.The conditions for producing linoleic acid isomerase were optimized by the response surface experiment.The results showed that the best carbon source and nitrogen source are glucose and beef extract respectively.The optimal conditions for producing linoleic acid isomerase are as follows:2%glucose,2%beef extract,C/N1∶1.5,cultivation time36h,cultivation temperature36.4℃,initial pH5.6.Under above conditions,the yield of linoleic acid isomerase reached1214.1U/mL,which was2.84times as high as that of the original strain.
