摘 要:为研究解淀粉芽孢杆菌HN(Bacillus amyloliquefaciens HN)固体发酵菌体的方法 ,本试验就固体发酵的不同底物(麸皮、玉米粉,豆粕,大豆粉),添加物质(碳源、氮源和无机盐类),料水比,发酵温度,发酵时间,干燥方法,激活特性等因子进行单因素试验和正交设计,探索菌体形成的最佳培养条件和使用方法。结果表明:在以麸皮为底物,温度28℃,料水比为1∶0.8,添加外源物质为可溶性淀粉0.2%,蛋白胨0.2%, KH2PO40.1%, MgSO4·7H2O 0.03%所形成的固体培养条件下进行48 h的发酵,解淀粉芽孢杆菌的菌数可达3.99×1010cfu/g以上,明显高于液体菌剂4.17×109cfu/mL。固体菌剂干燥过程中的菌数和含水量极显著相关(P <0.01),干燥后的菌剂再进行二次发酵,菌数可达3.72×1010cfu/g。In order to study the method for production of Bacillus amyloliquefaciens HN using solid state fermentation(SSF),the SSF conditions involved the substrate(wheat bran,corn flour,soybean meal and soybean flour),and additive(carbon sources,nitrogens and salts),the ratio of material and water,temperature,the fermentation time,drying method.The single-factor test,and orthogonal design were used to optimize the SSF medium of Bacillus amyloliquefaciens HN.The results showed that the number of Bacillus amyloliquefaciens HN using SSF was influenced by physiological and chemical nature of the agriculture by product.The optimized SSF conditions such as substrate wheat bran,the ratio of material and water 1∶0.8,temperature 28 ℃,fermentation time 48 h,0.2% soluble starch,0.2% peptone,0.1% KH2 PO4,0.03% MgSO4·7 H2 O were determined. When SSF was adopted the optimized conditions,the biomass concentration of Bacillus amyloliquefaciens increased to3.99×1010 cfu/g,more than the liquid fermentation 4.17×109 cfu/mL.There was a significant correlation between the number of bacteria and the water content of solid fermentation agent during drying(P < 0.01),the bacteria number of solid fermentation agent increased to 3.72×1010 cfu/g after second fermentation.
