摘 要:目的:研究力竭运动对大鼠骨骼肌肌纤维类型及AMPK、TSC2、m TOR的影响。方法:选取SD雄性大鼠24只,随机分为4组,每组6只。对照组(C)不运动,其余3组均进行一次性跑台力竭运动,并于力竭后分别于即刻、24小时、48小时后宰杀(即E0、E24、E48);各组分离血清,并取下右侧股四头肌,一分为二,一份保存待测AMPK、TSC2、m TOR的含量及变化,另一份立即用于冰冻切片,观察肌纤维类型。结果:(1)肌纤维类型ATP酶染色结果显示:骨骼肌Ⅱ型肌纤维呈黑褐色,I型肌纤维呈灰白色;相同视野下,I型肌纤维的横切面积明显小于Ⅱ型肌纤维横切面积,且力竭运动各组I型肌纤维百分比明显低于对照组(P <0. 01),而Ⅱ型肌纤维百分比明显高于对照组(P <0. 01);但力竭运动各组(E0、E24、E48)之间差异不显著。(2) ELISA结果显示:力竭运动即刻组骨骼肌、血清中AMPK、TSC2含量明显高于对照组(P <0. 01),而力竭后24h、48h组骨骼肌、血清中AMPK、TSC2含量较对照组无明显变化。另一方面,力竭运动即刻组骨骼肌、血清中m TOR含量则明显低于对照组(P <0. 01),而力竭后24h、48h组骨骼肌、血清中m TOR含量较对照组无明显变化。且E24组与E48组间均无显著性差异。结论:(1)力竭运动后及恢复期间,骨骼肌I型肌纤维百分比降低,Ⅱ型肌纤维百分比增加。(2)力竭运动即刻组的AMPK、TSC2含量明显高于其余各组,而m TOR含量则低于其余各组,提示力竭运动中AMPK可能被激活,同时磷酸化TSC2进而抑制m TOR的合成。其与肌纤维类型变化的关系有待进一步研究。Objective To investigate the effect of exhaustion exercise on the type of muscle fiber and AMPK,TSC2,mTOR in skeletal muscle of rats.Methods Twenty-four SD male rats were randomly divided into four groups(n=6)and control group(C)did not exercise.The exercise groups were given disposable treadmill exhaustive exercise and slaughtered after exhaustion instantly and 24 hours and 48 hours(E0,E24,E48).The blood serum were taken and the right quadriceps were divided into 2 parts.One was saved in a 80℃fridge to detect the contents of AMPK,TSC2,mTOR in all groups,and the other one was taken immediately for frozen section to observe the type of muscle fiber.Results(1)mATPase staining results of muscle fiber type showed that:Type II skeletal muscle fibers were dark brown and type I muscle fibers were grey.From the same visual field,the cross-sectional area of type I muscle fibers was significantly less than that of the type II muscle fiber,and the percentage of type I muscle fibers in all exhaustive groups was significantly lower than the one of the control group(P<0.01).On the other hand,the percentage of type II muscle fibers in all exhaustive groups was higher than that of the control group(P<0.01).But there were not significantly differences among all exhaustive groups.(2)The results of ELISA shows that:The contents of AMPK,TSC2 in skeletal muscle and in serum of E0 group were significantly higher than the one of the control group(P<0.01).But the contents of AMPK,TSC2 in E24 and E48 had no significantly change compared to the control group.On the other hand,the contents of mTOR in skeletal muscle and in serum of E0 group were obviously lower than that of the control group(P<0.01).But the ones of mTOR in E24 and E48 had no significantly change compared to the control group.And there were no significantly differences between E24 group and E48 group.Conclusions(1)The percentage of type I muscle fibers decreased and the percentage of type II muscle fibers increased during the exhaustive exercise and recovery.(2)The conten
