摘  要:目的:探讨肿瘤坏死因子-α(TNF-α)基因rs1800629位点与精神分裂症及其临床特征的关系。方法:采用Taq Man探针SNP基因分型技术对稳定期精神分裂症患者(患者组) 254例和健康对照者(正常对照组) 339名TNF-α基因rs1800629位点进行分型;采用阳性和阴性症状量表(PANSS)评定患者组精神病性症状,分析比较rs1800629位点3种不同基因型间症状严重程度。结果:患者组与正常对照组TNF-α基因rs1800629位点基因型的观察值和期望值Hardy-Weinberg平衡吻合度良好(χ2=2. 70,P=0. 10;χ2=2. 58,P=0. 11);患者组与正常对照组在rs1800629位点基因型和等位基因频率分布比较,差异无统计学意义(χ2=3. 16,P=0. 21;χ2=3. 44,P=0. 06);携带A等位基因患者阳性症状分、阴性症状分、一般病理分及PANSS总分较不携带A等位基因患者更高(F=6. 03,P=0. 015; F=14. 82,P=0. 000; F=42. 02,P=0. 000; F=35. 19,P=0. 000)。结论:TNF-α基因rs1800629位点可能与精神分裂症无关;但与疾病的严重程度有关。Objective:To investigate the relationship between rs1800629polymorphism in TNF-αgene and schizophrenia.Method:We recruited254schizophrenia patients with stable condition(case group)and339healthy controls(control group).TaqMan technique was carried out to genotype rs1800629polymorphism.Positive and negative syndrome scale(PANSS)was used to evaluate the psychotic symptoms in patients with schizophrenia,and analyze the symptom severity among three genotypes of rs1800629polymorphism.Results:No deviation from the Hardy-Weinberg equilibrium was observed in rs1800629genotype distribution(χ^2=2.70,P=0.10;χ^2=2.58,P=0.11).There were no significant differences in genotypic or allelic frequencies of the rs1800629polymorphism between the case and control groups(χ^2=3.16,P=0.21;χ^2=3.44,P=0.06).Patients with A allele had higher scores in positive symptoms,negative symptoms,general psychopathology and total score than those without A allele(F=6.03,P=0.015;F=14.82,P=0.000;F=42.02,P=0.000;F=35.19,P=0.000).Conclusion:This study did not support TNF-αgene conferring susceptibility to schizophrenia,however,suggested that this gene may contribute to severity of clinical symptoms.
