摘 要:【目的】探究不同孔径陶瓷膜澄清处理罗非鱼鱼肉蛋白酶解液的澄清效果。【方法】考察200 nm和50 nm陶瓷膜的膜通量、透过液理化特性随处理时间的变化以及膜的清洗条件,采用凯氏定氮法与福林酚法分析蛋白质含量,全自动色差计测定色差值。【结果】在批处理模式(TBM)、循环流量20L/min和跨膜压力差0.06MPa条件下,200 nm和50 nm陶瓷膜的膜通量均于操作11 min后达到平稳,分别为91.0 L/(m2·h)和71.0 L/(m2·h);透过液中可溶性蛋白质含量于操作10 min后达到平稳,蛋白质回收率分别为87.58%与85.84%;陶瓷膜可明显降低酶解液的色差值,脱色澄清效果较显著,且50 nm比200 nm的效果好;采用质量分数为0.5%HNO3和0.5%NaOH分步清洗方法,200 nm和50 nm的陶瓷膜的膜通量可分别恢复92.56%和88.75%。【结论】陶瓷膜过滤适合用于罗非鱼肉蛋白酶解液澄清处理。【Objective】To investigate the clarification effect of different pore sizes ceramic membrane on tilapia meat protein hydrolysates.【Methods】The objectives of this study were to investigate the effects of membrane flux of 200 nm and 50 nm ceramic membrane, to analyze the physical and chemical properties of permeate liquid with the change of micro filtration time and membrane cleaning condition. The protein of hydrolysate was assayed by kjeldahl and folin-phenol method. The color difference value was assayed by automatic color difference meter. 【Results】Using TBM, circular flow of 20 L/min and transmembrane pressure difference of 0.06 MPa were applied, the membrane flux of 200 nm and 50 nm was stable after 11 min of operation and was 91.0 L/(m2·h) and 71.0 L/(m2·h), respectively. Soluble protein content of permeate liquid was stable after 10 min of operation, nitrogen recovery rate was 87.58% and 85.84% respectively. Ceramic membrane can significantly clarify and reduce the color difference value of hydrolysates and the effect of 50 nm was better than that of 200 nm. Using 0.5% HNO3 and 0.5% NaOH to clean up step by step, the recoveries of ceramic membrane flux of 200 nm and 50 nm were 92.56% and 88.75%, respectively. 【Conclusion】Ceramic membrane was fit for clarifying and decoloring of tilapia meat protein hydrolysates.
