摘  要:葡萄皮干燥磨粉,经水浸法提取、乙醇沉淀、石油醚脱脂、Sevage脱蛋白方法提取多糖,多糖用三氟乙酸水解,衍生化处理后,采用反相高效液相色谱-紫外检测法分析检测。经研究,适合PMP衍生物分离的色谱柱为TC-C_(18),流动相为0.05 mol/L磷酸盐缓冲液-乙睛,最佳柱温为25℃,适宜梯度洗脱流速为1 mL/min。结果显示,5种单糖的含量为甘露糖0.114 7 mg/g、鼠李糖1.290 7 mg/g、半乳糖醛酸0.346 5 mg/g、半乳糖3.9514 mg/g、阿拉伯糖3.127 9 mg/g。Grape peel dry grinding powder was extracted by water immersion,ethanol precipitation,petroleum ether degreasing and Sevage deproteinization.Polysaccharides were hydrolyzed by trifluoroacetic acid and then derivatized.The polysaccharides were analyzed by reversed-phase high performance liquid chromatography-ultraviolet detection.The optimum column temperature and gradient elution flow rate for PMP derivatives were 25℃,1 mL/min and TC-C18,0.05 mol/L mobile phase phosphate buffer-acetonitrile.The results showed that the contents of five monosaccharides were mannose 0.114 7 mg/g,rhamnose 1.290 7 mg/g,galacturonic acid 0.346 5 mg/g,galactose 3.951 4 mg/g and arabinose 3.127 9 mg/g.
