摘  要:建立了高效液相色谱法测定大蒜愈伤组织中大蒜素含量的分析方法。色谱柱为Eclipse Plus C18(4.6mm×250mm,5μm),柱温35℃,流动相为甲醇-水含0.2%甲酸(80∶20),流速1.0mL/min,二极管阵列检测器检测波长240nm,进样量50μL。结果显示:大蒜素在0.30~4.57μg/mL范围内线性关系良好,相关系数R2=0.999,加标回收率为95%~102%。该方法灵敏度高,重现性好,操作简便,适用于大蒜愈伤组织中大蒜素含量的检测。Develop a method for the determination of allicin in garlic callus by high performance liquid chromatography(HPLC).The conditions of HPLC are as follows:the chromatography column is Eclipse Plus C 18(4.6 mm×250 mm,5μm),the column temperature is 35℃,the mobile phase is methanol∶water(0.2%formic acid)of 80∶20(V/V),the flow rate is 1.0 mL/min,the DAD detector wavelength is 240 nm,and the sample injection volume is 50μL.The results show that the content of allicin has a good linear relationship in the range of 0.30~4.57μg/mL,the correlation coefficient is 0.999,and the average recovery of allicin is 95%~102%.This method is accurate,reliable,simple and efficient for the detection of allicin in garlic callus.
