摘  要:采用光谱和酶动力学方法研究青砖茶粗多糖对α-葡萄糖苷酶活性的抑制作用及机制。结果表明:青砖茶粗多糖对α-葡萄糖苷酶的抑制作用接近于阳性对照品阿卡波糖,二者的半抑制浓度依次为(2.04±0.07)mg/m L和(2.00±0.03)mg/m L,不存在显著性差异(P>0.05);Lineweaver-Burk双倒数作图显示青砖茶粗多糖的抑制类型为竞争性抑制,且紫外吸收光谱实验表明青砖茶粗多糖与α-葡萄糖苷酶之间存在相互作用。文章为深入开展青砖茶粗多糖降血糖活性研究及其功能性产品开发提供了理论基础。The inhibitory effects of crude polysaccharide of green brick tea on α-glucosidase were investigated by spectroscopy and enzyme kinetics, and the possible mechanisms involved were explored.The results showed that the inhibition activity of crude polysaccharide of green brick tea was close to that of acarbose as a positive control, and the half inhibitory concentration(IC50) values of crude polysaccharide and acarbose were(2.04±0.07)mg/mL and(2.00±0.03)mg/mL, respectively, and there is no significant difference between their IC50 values(P>0.05). The Lineweaver-Burk double reciprocal plot indicated that the inhibition type of crude polysaccharide of green brick tea was competitive inhibition,and Ultraviolet absorption spectra showed that there is interaction between crude polysaccharide of green brick tea and α-glucosidase. The results of the study provided the basis for further research on hpyerglycemic activity of crude polysaccharide of green brick tea as well as their functional product development.
