摘 要:目的探讨大豆肽(soybean peptide,SP)通过下调Notch 1信号通路防治哮喘大鼠气道炎症反应的作用机制。方法将60只SD雄性大鼠随机分为正常对照组、哮喘模型组、地塞米松(Dexamethasone,Dex)0.5 mg/kg组及大豆肽600、300及150 mg/kg剂量组。采用卵清蛋白(Ovalbumin,OVA)+氢氧化铝(Aluminum hydroxide,Al(OH)_3)法复制大鼠哮喘动物模型,同时给予相应剂量的大豆肽进行干预,每天给药1次,连续7 d。末次给药24 h后收集大鼠肺泡灌洗液(broncho-alveolar lavge fluid,BALF),进行白细胞分类计数;取大鼠右下叶肺组织常规HE染色,观察肺组织病理学改变及气道重塑变化;采用逆转录PCR和Western-blot法检测大鼠肺组织Notch 1、Jagged 1、Hes 1及NF-κB mRNA和蛋白表达水平。结果与哮喘模型组比较,大豆肽600 mg/kg剂量组大鼠BALF中的中性粒细胞、淋巴细胞及嗜酸性粒细胞计数显著降低(P<0.01);大鼠肺组织病理损伤较模型组明显改善,炎性细胞浸润明显减少;大鼠肺组织Notch 1、Jagged 1、Hes 1及NF-κB mRNA和蛋白表达水平明显下调(P<0.05,P<0.01)。结论大豆肽对哮喘大鼠炎症反应具有防治作用,其机制与下调Notch 1/Jagged 1/Hes 1/NF-κB信号通路有关。Objective To observe the inhibition inflammation effect of soybean peptide ( SP) on the airway of rats with bronchial asthma, and to explore its underling mechanism about Notch 1 signaling pathway. Methods Sixty SD rats were divided into normal control group,asthma model group,dexamethasone 0. 5 mg /kg group,and soybean peptide 600,300 and 150 mg /kg group,randomly. The asthma rat model was established by ovalbumin + Al ( OH) 3 sensitization,and the different dose of SP were given the model rats, once a day for 7 days. After 24 h in the last administration, the bronchoalveolar lavage fluid ( BALF) was collected to detect the classified and counter of leucocyte. The pathological changes of lung tissue and airway remodeling were observed by H&E staining. The mRNA and protein expression levels of Notch 1,Jagged 1,Hes 1and NF -κB of lung tissue were analyzed and determined by reverse transcription - polymerase chain reaction ( RT - PCR) and Western blotting methods. Results Compare with the asthma model group, the SP ( 600 mg /kg) could decrease the neutrophil, lymphocyte and eosinophils in BALF of asthma rats ( P < 0. 01), relief the pathological damage of lung tissue and down - regulate the mRNA and protein expression levels of Notch 1,Jagged 1,Hes 1 and NF -κB of lung tissue ( P < 0. 05,P < 0. 01). Conclusion SP has the effect of anti - inflammation in asthma rats,and its mechanism may be related to down - regulate the Notch 1 /Jagged 1 /Hes 1 /NF -κB signaling pathway.
