摘  要:通过柱层析技术从诸葛菜种子中分离抗真菌蛋白,并采用纸片扩散法、琼脂平板稀释法和凹玻片法对抗真菌蛋白的抑菌活性和稳定性进行研究.结果表明:从诸葛菜种子中分离出一种分子质量约为10 ku的抗真菌蛋白,命名为Zp;经QE质谱鉴定,Zp蛋白可能是诸葛菜种子中的一种防御素蛋白.Zp蛋白对11种植物病原真菌均具有较强抑制作用,其中对尖孢镰刀菌生长的半抑制浓度(IC_(50))为54.36μg·mL^(-1),对其孢子萌发的IC_(50)为15.97μg·mL^(-1).Zp蛋白的抑菌活性在温度0~100℃和pH 1~14的范围内稳定,其对有机溶剂(15%甲醇、乙醇、异丙醇、乙醚和丙酮)和金属阳离子(50~250 mmol·L^(-1) Mg^(2+)、K^+、Mn^(2+)和Ca^(2+))也具有较强的耐受力.An antifungal protein was isolated from Orychophragmus violaceus seeds by column chromatography,and its antifungal ac tivity and stability were investigated by paper diffusion,agar plate dilution and concavity slide methods.The protein purified was named Zp,it was 10 ku and identified as a defensin protein by mass spectrometry.Zp protein had strong inhibitory effects on 11 plant pathogenic fungi.The IC50 values of Zp protein against the growth and spore germination of Fusarium oxysporum were 54.36 and 15.97μg·mL-1,respectively.Its antifungal activity was stable between 0 and 100℃and under pH 1-14.It also exhibited high re sistance to organic solvents(15%methanol,ethanol,isopropanol,ether and acetone)and metal ions(50-250 mmol·L-1 Mg2+,K+,Mn2+and Ca2+).
