摘 要:采用响应面分析法优化荔枝多糖超声-微波提取的最佳工艺条件;分析小鼠器官指数、血清溶血素、NK细胞活性、脾淋巴细胞增殖,探究荔枝多糖体内免疫调节活性。结果表明,荔枝多糖超声-微波的最佳提取工艺条件为p H 8.2,微波功率559 W,提取时间10.8 min,水/料比22.7∶1,在此提取条件下,多糖提取产率可达24.02%,高于传统的热水法、微波法和超声法。体内免疫调节活性评价实验表明:在50~200 mg/kg范围,荔枝多糖均能提高小鼠的血清溶血素水平、胸腺器官指数、NK细胞杀伤活性以及促进脾淋巴细胞增殖,且中剂量(100 mg/kg)荔枝多糖的免疫活性最强。由此表明,采用超声-微波法能显著提高荔枝多糖的提取率,且获得的荔枝多糖具有显著的体内免疫调节作用。Response surface analysis experiment was carried out to optimize the polysaccharides extraction from litchi pulp.The immunomodulatory effects of obtained litchi polysaccharides (LP) were further evaluated in normal KM mice.The results showed that the optimal extraction conditions were as follows :pH 8.2, power 559 W, extraction time 10.8 min, water/solid ratio 22.7 :1 and the yield was 24.02% under this condition, which was significantly higher than those of hot extraction, ultrasonic -assisted extraction and microwave -assisted extraction.In addition, the administration of LP (5 0 ,1 0 0 mg/kg and 200 mg/kg) remarkably enhanced serum hemolysin production, increased the thymus index, NK cells cytotoxicity and splenocyte proliferation in KM mice.Middle dose group (100 mg/kg) LP exhibited the best immunomodulatory activities among three doses.In summary, the ultrasonic-microwave assisted extraction significantly increased the yield of litchi polysaccharide and obtained litchi polysaccharides exerted their immunomodulatory activities in vivo.
