摘 要:目的:通过生物信息学方法分析微管相关蛋白tau(MAPT)在乳腺癌中的表达及其临床意义。方法:分别从TCGA、bc-GenExMiner、GEO、Kaplan Meier Plotter等公共数据库当中下载乳腺癌中MAPT表达的数据资料,分析MAPT在乳腺癌中的表达情况及其与乳腺癌类型、治疗、预后的关系。结果:总体分析结果显示,MAPT在乳腺癌组织中表达高于正常乳腺组织(P<0.01),但分组分析显示,MAPT在恶性程度低、临床分期早、内分泌治疗敏感的乳腺癌中呈高表达,反之则相反。MAPT高表达乳腺癌对他莫昔芬敏感性更高但容易对紫杉醇耐药(P<0.01)。总体分析结果显示,MAPT高表达乳腺癌患者的总生存期与无复发生存期均长于其低表达乳腺癌患者(HR=0.65,95%CI=0.48~0.89,P<0.05;HR=2.91,95%CI=1.54~5.51,P<0.05),但分组分析显示,仅LuminalA型患者与总体结果一致,其余类型或无关或相反。结论:MAPT的表达在不同类型的乳腺癌中存在差异,且与乳腺癌的耐药与预后密切相关,在指导乳腺癌治疗和评估预后方面具有一定临床意义。Objective: To analyze the expression of microtubule-associated protein tau (MAPT) in breast cancer and its clinical significance though bioinformatics approach. Methods: Data about MAPT expression in breast cancer were downloaded from the public databases that included TCGA, bc-GenExMiner, GEO and Kaplan Meier Plotter. The MAPT expression profiles and their relations with the types of breast cancer as well as the treatment and prognosis of breast cancer were analyzed. Results: The result of overall analysis showed that MAPT expression was higher in breast cancer tissue than that in normal breast tissue (P<0.01), but the results of stratified analysis showed that MAPT expression was increased in breast cancer with low malignancy, early clinical stage or sensitivity to endocrine therapy and conversely, the opposite was true. Breast cancer with high MAPT expression was more sensitive to tamoxifen but more likely resistant to paclitaxel (P<0.01). The result of overall analysis showed that the overall survival and relapse-free survival in patients with high MAPT expression were longer than those in patients with low MAPT expression (HR=0.65, 95% CI=0.48–0.89, P<0.05;HR=2.91, 95% CI=1.54–5.51, P<0.05), but the results of stratified analysis showed that consistent result was only achieved in patients with Luminal A breast cancer, while the irrelevant or opposite results were seen in patients with other types of breast cancer. Conclusion: MAPT expressions are diversified in different types of breast cancer, which were closely associated with drug resistance and prognosis of breast cancer. So, it is of certain clinical significance for treatment selection and the prognostic assessment of breast cancer.
