摘 要:为了解不同消毒剂对大黄鱼中致病性弧菌的抑菌效果。采用纸片扩散法比较了聚维酮碘(PVP-I)、聚六亚甲基双胍(PHMB)和过氧化氢(H2O2)对溶藻弧菌和副溶血性弧菌的抑菌效果,再以肉汤稀释法测定了三种消毒剂的最低抑菌浓度(MIC)、最低杀菌浓度(MBC)及MBC/MIC比值,在此基础上,获得了抑菌效果相对较优的H2O2、PHMB对溶藻弧菌、副溶血性弧菌及其混菌的杀菌动力学曲线。结果表明,H2O2(3%)对两种弧菌具有最大的抑菌圈直径(19~21mm),PHMB(30mg/mL)次之(约为15mm),PVP-I(10%)的抑菌效果最弱(小于10mm)。3种消毒剂的MIC值由小到大依次为PHMB<H2O2<PVP-I,MBC/MIC均小于等于2。时间杀菌曲线表明,1MIC时,H2O2和PHMB处理均表现为杀菌作用。1MIC的H2O2在2h、1h、2h内已完全杀灭溶藻弧菌、副溶血性弧菌和混合弧菌,且作用持久。1MIC的PHMB则在1h达到了完全杀灭作用,杀菌速度相对较快。本研究对大黄鱼致病弧菌的防治药物选择及使用剂量的确定可提供参考。In order to observe the antibacterial activity of different disinfectants on pathogenic vibrios of Pseudosciaena crocea , the antibacterial effects of PVP-I, PHMB, H 2O 2 on Vibrio alginolyticus and Vibrio parahaemolyticus were evaluated by disc diffusion method. The MIC, MBC and MBC/MIC values of these three disinfectants on the two vibrios were obtained by broth microdilution method. Furthermore, the time-kill curves of H 2O 2 and PHMB against Vibrio alginolyticus, Vibrio parahaemolyticus and mixed strain of these two vibrios were also determined. The results showed that, with the optimal concentration, H 2O 2 had the biggest diameter of inhibition zone (19~21 mm) for both vibrios, followed by PHMB (about 15 mm), while PVP-I had the weakest bacteriostatic effect (less than 10 mm). Based on the value of MIC, it could be ranked with the order of PHMB<H 2O 2<PVP-I. However, all the MBC/MIC values were less than or equal to 2. The time-kill assay results confirmed that H 2O 2 and PHMB showed bactericidal activity. At the concentration of 1 MIC, Vibrio alginolyticus, Vibrio parahaemolyticus and mixed bacteria of both vibrios were killed completely by H 2O 2 within 2 h, 1 h, 2 h, respectively, and kept inhibited. The complete killing time of PHMB was no more than 1 h, which was much faster than that of H 2O 2. In conclusion, the results of this study would provide a reference for the selection of the disinfectants and the dosage for effectively controlling the pathogenic vibrios of Pseudosciaena crocea .
