摘 要:【目的】优化太空茄子叶中总黄酮的提取工艺.【方法】以太空茄子叶为试材,在单因素试验的基础上,取乙醇体积分数、料液比、提取时间为自变量,太空茄子叶总黄酮得率为响应值,以响应面法得到二次多项式模型,优化其提取条件,并采用HPLC对总黄酮进行分析和鉴定.【结果】在温度(80 ℃)固定的条件下,太空茄子叶总黄酮的最佳工艺条件为:提取时间1.8 h、料液比1 ∶ 42(g/mL)、乙醇体积分数78%,在此条件下,太空茄子叶总黄酮得率为9.82%,与预测值9.87%相吻合.【结论】响应面分析法优化太空茄子叶总黄酮的提取工艺合理可行.【Objective】 To optimize the extraction process of total flavonoids from space eggplant leaves.【Method】 On the basis of a single factor experiment,space eggplant leaves were used as the test material,ethanol concentration,solid-to-liquid ratio,and extraction time were used as independent variables,the extraction yield of total flavonoids was used as response value.A quadratic polynomial model was established according to the response surface methodology.By optimizing the extracting condition,the total flavonoids were analyzed and identified by HPLC.【Result】 At the constant ambient temperature of 80 ℃,the optimal conditions for ethanol reflux extraction of total flavonoids were found to be as follows,extraction time 1.8h,solid-to-liquid ratio 1 ∶ 42(g/mL),ethanol concentration 78%.Under these conditions,the extraction yield of total flavonoids was 9.82%,which was fully in agreement with the predicted value of 9.87%.【Conclusion】 It is suggested that response surface analysis to optimazation of extraction process of total flavenoids from space eggplant leaves is feasible.
