摘  要:目的本实验对大豆中低聚糖的分析方法进行研究,比较非转基因和转基因大豆中低聚糖的含量。方法样品用乙腈水溶液溶解,经超声提取、离心过膜后,超高效液相-示差折光分析仪(UPLC-RI)测定,外标法定量。结果本实验应用40%乙腈水直接对大豆样品进行溶解超声提取,方法的回收率为91%~97%,相对标准偏差小于5%。非转基因和转基因大豆样品的低聚糖分析,转基因大豆中的含量与非转基因大豆不存在显著差异,可以满足人体健康需要。结论该方法准确快速、重现性好,适用于大豆中低聚糖含量分析的检测要求。Objective In this study, the method for determination of oligosaccharides in soybean was established, and the contents of oligosaccharides in non-transgenic and transgenic soybeans were compared. Methods The samples were treated with to acetonitrile aqueous solution, ultrasonic extraction and centrifugation, and were detected by the ultra high performance liquid chromatograph with refractive index (UPLC-RI). The external standards were used for quantitative analysis. R e su lts The 40% acetonitrile water was used to directly extract soybean samples by solvent ultrasonic extraction. The recovery rate of the method was 91%-97%, and the relative standard deviation was less than 5%. Through the analysis of oligosaccharide contents no significant difference was found between transgenic and non-transgenic soybeans. Conclusion The method was accurate of good reproducibility and suitable for determination of igosaccharides in soybeans.
