摘  要:研究了近平滑假丝酵母ATCC7330的高压均质破碎工艺,利用正交试验考查了均质温度、酵母质量分数、均质压力和均质次数对酵母细胞破碎得到的酮基还原酶的酶活影响。结果表明,4个因素的影响大小依次为均质压力>酵母质量分数>均质温度>均质次数,其中均质压力为近平滑假丝酵母破碎的显著性因素,最佳破碎工艺为均质温度6℃,酵母质量分数20%,均质压力180MPa,均质次数为3次。经过验证试验,采用高压均质破碎酵母细胞的方法可得到粗酶液酶活1.572U/mL,比优化前提高150%以上。The breaking process of Candida parapsilosis ATCC 7330 through high pressure homogenization was investigated. The effect of the temperature,the yeast concentration,the pressure,the times of the homogenization on the ketoreductase activity was studied. The results showed that the effect order was the pressure > the yeast concentration > the temperature > he times during the homogenization and the pressure was the obvious factor for the Candida parapsilosis ATCC 7330 breaking process. The optimal breaking process was obtained for the temperature 6 ℃,the yeast concentration 20%,the pressure 180 MPa and recycling three times. The crude ketoreductase activity was 1.572 U/mL,increasing 150% before the optimization.
