摘  要:以碳酸钙微球作为模板剂、多巴胺为包覆剂,制备氮掺杂多孔碳材料,并将其用于木瓜蛋白酶的固定化。优化的固定化条件为pH6.5、酶浓度为4g/L和固定化温度为50℃,测得酶的固定化量为1614.37mg/g,酶最大的比活为2.22×10^4U/g。与游离酶相比,固定化木瓜蛋白酶显示出提高的pH稳定性、热稳定性和储存稳定性。在70℃的温度条件下放置1h后,固定化木瓜蛋白酶依然保留其初始活性的56.7%,而游离木瓜蛋白酶仅为11.5%。储存10d后,固定化木瓜蛋白酶仍保留了其初始活性的72.9%,而游离蛋白酶在第10天时只剩余5%的活性。在重复使用5次后,固定化木瓜蛋白酶仍具有较高的酶活力。因此氮掺杂多孔碳材料作为固定化酶载体具有良好的应用前景。CaCO3 microspheres and dopamine were used as template and coating agent respectively to prepare hollow N-doped carbon materials (HNCM),and then HNCM as a carrier for immobilizing papain. The optimized immobilization condition was pH 6.5,papain concentration was 4 g/L and immobilization temperature was 50 ℃,the immobilized amount of the papain was 1614.37 mg/g and maximum papain activity was 2.22×10^4 U/g.Immobilized papain showed pH stability,thermal stability and storage stability than free papain.After 1 h incubation at 70 ℃,the immobilized papain retained 56.7% of its initial activity while the free papain only retained 11.5%.The immobilized papain maintained 72.9% of their initial activity after being stored for 10 days,while the free papain maintained only 5%.Furthermore,immobilized papain still has high activity after five consecutive operations.Therefore,hollow N-doped carbon materials have good application prospects as immobilized enzyme carriers.
