摘 要:为了探索杜仲皮水提物对虹鳟肌肉生肌调节因子家族(myogenic regulator factors, MRFs)的调节作用,采用荧光定量PCR法对其mRNA表达量进行检测分析。本研究选取初始体重为(145.56±4.12) g的虹鳟进行试验,设置5个处理组,即在基础饲料中分别添加质量分数为0%(对照组)、0.5%、1.0%、2.0%和4.0%杜仲皮的水提物,养殖10周。结果显示,杜仲皮水提物对虹鳟肌肉各处理组之间Myf6、MyoD和Myf5基因表达量差异不显著(P>0.05);但是对MyoG基因表达量影响较为明显,其中2.0%组肌肉MyoG基因表达量显著高于各处理组(P<0.05),4.0%组肌肉MyoG基因表达量显著高于对照组、0.5%组和1%组(P<0.05)。杜仲皮水提物可通过调控虹鳟肌肉MyoG基因的表达以实现肉质改善。本研究结果为进一步深入探究杜仲改善养殖动物肌肉品质的分子机理提供了一定的理论依据。In order to study the effect of the wa. ter extract of Eucommia ulmoides Oliver on the family of the MRFs gene in the muscle of rain. bow trout, the mRNA expression of the muscle tissue was analyzed by the fluorescence quantita. tive PCR from the molecular level. The fish with an initial body weight of (145.56±4.12) g were fed one of the five diets for 10 weeks, including 0 g/kg (control, 0%), 5.00 g/kg (0.5%), 10.00 g/kg (1.0%), 20.00 g/kg (2.0%) and 40.00 g/kg (4.0%) barks extract. The results showed that there were no significant differences in the expression of Myf6, MyoD and Myf5 genes among all the treatments (P>0.05). However, the expression of MyoG gene in 2.0% group was significantly higher than that of the other groups (P<0.05), and the expression of MyoG gene in 4.0%group was significantly higher than that of control group, 0.5% group and 1%group (P<0.05). The water extract of Eucommia ulmoides could regulate the expression of MyoG gene in muscle of rainbow trout.The results of this study provide a basic theory for further exploring the molecular mechanism of E. ulmoides Oliver in improving muscle quality of cultured animals.
