摘  要:采用拉曼光谱、红外光谱、圆二色性光谱和X-射线衍射法测定了丝素蛋白经氯化钙溶液溶解前后的结构。结果表明,精练丝素的结构以β-折叠为主,存在晶区;可溶性丝素粉末和丝素溶液以无规卷曲为主,基本呈无定形状态。氯化钙溶解丝素的机理可能是,主要通过氯化钙浓溶液的强水化作用、氯离子和钙离子与丝素中酪氨酸残基的结合破坏了丝素分子间的范德华力和氢键,从而使丝素发生膨润而溶解。The structure of silk fibroin before and after dissolving with CaCl_2 solution was detected by Raman spectroscopy, Infrared (IR) spectroscopy, circular dichroism (CD) and X-ray diffraction. Results showed that the refining fibroin was in the β-sheet structure and contained crystal structure. When treated with CaCl_2 solution, the β-sheet structure mainly changed to random coil, and the crystal structure mainly changed to amorphous. The mechanism of dissolving fibroin with CaCl_2 solution has been deduced as follows. Because of the high hydration of the concentrated CaCl_2 solution, and the ability of calcium and chloride ions to bind with tyrosine residues in fibroin, the van der Waals force and hydrogen bonds of fibroin molecules are disrupted. This will cause fibroin to swell and then dissolve.
