摘 要:目的 研究乙型肝炎病毒 (HBV)S区蛋白与环孢素A(CsA)之间的肝细胞毒性关系 ,以探讨HBsAg阳性的肾移植患者使用CsA后 ,肝炎活动发生率显著增高的可能机制。方法 建立了稳定表达HBVS区蛋白的肝癌细胞株 (HepG2 )细胞克隆 ,并以此为模型 ,用MTT试验及DNAfragmen tation等方法 ,观察HBVS区蛋白在细胞内的表达对CsA及Fas单抗 (FasAb)诱导HepG2细胞凋亡的影响。结果 单独使用CsA (1μg/ml)对HepG2细胞并无毒性效应 ,但在FasAb的协同下可诱导HepG2细胞凋亡。HBVS区蛋白对CsA及FasAb的这一作用无影响。 结论 CsA与FasAb的协同细胞毒性效应 。Objective To elucidate the possible pathogenesis for HBsAg-positive patients with high risk in active hepatitis when treated with cyclosporin (CsA) and the relationship between the cellular expression of envelope proteins of hepatitis B virus (HBV) and the cytotoxicity of CsA.Methods The cells expressing envelope proteins stably were established and used as model. The effects of envelope proteins of HBV on HepG2 undergoing apoptosis induced by CsA were investigated by MTT assay and DNA fragmentation experiment. Results CsA used alone had no apparent toxicity to HepG2, but in the presence of of Fas antibody (FasAb) it could induce marked apoptosis in HepG2 cells. This cytoxicity was not affected by envelope proteins of HBV. Conclusion The synergic effect of CsA and FasAb on cytotoxicity might be related to the increased risk of hepatic dysfuction in patients infected with HBV.
